ImageQuant LAS 4000

1. メンブレンをハイブリバックに入れ、空気を抜き、シールする。あるいは、サランラップを敷き、メンブレンの表を下にして乗せ、サランラップで閉じる。
2. CD-Rを持参し、ImageQuant LAS 4000 mini or ImageQuant LAS 4000、パソコンを起動する。
3. ImageQuant LAS 4000のソフトを起動する。
4. メンブレンをトレイにセット
5. Method/Tray position→Chemiluminescence→Tray position→OK。
6. Focusingで焦点を確認。
7. Exposure TypeをIncrement→1min→Sensitivity/ResolutionをStandard→Start
8. 撮影が終わったら、Stop→Save。
9. Save selected images→16枚まで保存
10. Save passed images→保存
11. (明視野と重ね合わせる場合は)Exposure TypeをPrecision→長さを設定→Add Digitization ImageをON→START→SAVE→Next
12. ImageQuant TLを起動→Analysis Toolbox
13. 画像を開く→Contrast変更→Edit→Export to File

明視野と重ね合わせる場合
14. File→Create Multiplex Image→名前入力→Browse→Ctrlを押しながら画像2つを選択→open→create→Colour→Channel 1, 2をGray Scale
15. Edit→Export to File→OK→Save

バンドを定量する場合
14. 画像を開く→Edit Image→Freeform rotateで画像を回転→バンドの列を囲む→Image→Crop to area→File→Save as→画像を閉じる
15. 切り取ったバンド画像を開く→Areas→バンドを拡大し、囲む→Next→background shape→背景を囲む→Selector→バンドを選択→Image Rectangle/Ellipse→Next
16. Area windowをactiveにし、edit→Export to file→Volumeの値を解析する。
